Effect of Introducing Perilla frutescens (L.) Britt DNA on Rutin Content in Tobacco Leaves
This paper studied the content of rutin in the progenies of fluecured tobacco with introduced exogenous DNA of
Perilla frutescens
(L.) Britt. The method of sexual hybridization combined with asexual graft was used to introduce exogenous DNA into tobacco
and the optimum environment of pollination was selected. HPLC was adopted to analyze the content of rutin in tobacco leaves. This method could significantly improve the success ratio of introducing exogenous DNA under the condition of lower temperature (22~25 ℃) and higher relative humidity (70 %~80 %). The results of the analysis showed that the content of rutin in the progenies increased by 70.4 % compared with that in female parent tobacco leaves and was 4~5 times higher than that in buds of
Perilla frutescens
L. from main production area in China. The progenies of fluecured tobacco with introduced exogenous DNA could provide new materials for massextraction of rutin. This study also explored a new way for utilizing gene resources in medicinal plants.
